Immunocytochemistry analysis of HepG2 cells labeling KRT15 with KRT15 Antibody (HY-P84150) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with KRT15 Antibody (HY-P84150) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L(HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Function
Cytokeratin 15 (CK15), also known as keratin-15 (KRT15), has been described as a stem cell marker based on its expression in human hair follicles and other organs. In animal models, CK15 has been shown to be a marker expressed in self-renewing, regenerative progenitor stem cells. In the breast, CK15 expression was reported in resting undifferentiated ductal luminal cells, with evidence linking CK15 expression with immature and basal-like properties. As for human cancers, CK15 expression is associated with aggressive tumour features and poor clinical outcomes in endometrial and oesophageal cancers[1].

Expressed in a discontinuous manner in the basal cell layer of adult skin epidermis, but continuously in the basal layer of fetal skin epidermis and nail.
