Background

Function
Fas receptor for TNFSF6/FASLG. The adapter molecule FADD recruits caspase CASP8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs CASP8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both. The secreted isoforms 2 to 6 block apoptosis (in vitro)

Expression
Tissue_specificity:Subtype 1 and subtype 6 were expressed at equal levels in resting peripheral blood mononuclear cells. Upon activation, subtype 1 levels increased, while subtype 6 levels decreased. -

Subunit
Component of the death-induced signaling complex (DISC) composed of cell surface receptor FAS/CD95, adapter protein FADD and the CASP8 protease; recruitment of CASP8 to the complex is required for processing of CASP8 into the p18 and p10 subunits (PubMed:21109225, PubMed:9184224, PubMed:9322534).
