  • Experimental Validation Results for STAT6 Antibody (YA5933)
    Western blot analysis was performed on protein extracts (25 μg) from HepG2 (lane 2), MCF-7 (lane 3), 293T (lane 4,Negative), HeLa (lane 5), HT-1080 (lane 6), and A549 (lane 7) using STAT6 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. Thhe primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:2500) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit IgG (H&L) secondary antibody (HY-P8001) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

    STAT6 is a cytokine-responsive transcription factor that links receptor signaling to gene regulation in immune cells[1]. Mechanistically, IL-4 and IL-13 activate STAT6, and this axis drives TH2 polarization, IL-4-dependent lymphocyte responses, and IgE class switching[1][2][3]. In disease models, STAT6 regulates pathologic lung inflammation and supports experimental analysis of allergic asthma, airway hyperresponsiveness, and atopic inflammation[4][5][6]. Compared with related STAT isoforms, STAT6 shows a distinct functional focus on IL-4/IL-13 biology, whereas STAT4 primarily controls TH1 differentiation[7]. For experimental applications, STAT6 inhibition with AS1517499 reduced antigen-induced bronchial hypercontractility and blocked STAT6 signaling in mouse atopic dermatitis-asthma models[5][6].
