Function
Membrane-associated guanylate kinase CASK is a multidomain MAGUK protein with Ca²⁺/calmodulin-kinase, PDZ, SH3, and guanylate kinase domains, and it localizes to neuronal synapses[1]. Mechanistically, CASK scaffolds synaptic protein complexes through interactions with neurexin, Veli/LIN-7, Mint 1, Caskin 1, syndecan-2, and PMCA4b[1][2][3][4]. CASK also links membrane signaling to nuclear regulation, because PMCA4b binding reduced T-element-dependent transcriptional activity in a Ca²⁺-dependent reporter system[4]. In disease models, CASK deletion in mice caused neonatal lethality and altered spontaneous glutamatergic and GABAergic release, while missense mutations associated with neurodevelopmental disorders disrupted binding to SAP97, CINAP, Tbr1, and neurexin[5][6]. Compared with related MAGUK proteins, CASK is distinguished by its Ca²⁺/calmodulin-dependent kinase domain and by alternative transcript variants that change binding affinities to interaction partners[5][7]. For experimental design, current literature supports CASK mutation, knockdown, knockout, splicing, and protein-interaction assays, but it does not establish validated CASK agonists or inhibitors for pathway-specific intervention[5][6][7].
