Background

Function
Matriptase-2 is a Membrane-bound serine protease. Through the cleavage of cell surface hemojuvelin (HJV), a regulator of the expression of the iron absorption-regulating hormone hepicidin/HAMP, plays a role in iron homeostasis[1][2][3][4][5].

Isoforms & Post-Translational Modification
Matriptase-2 has 4 isoforms, Q8IU80-4: amino acid length is 811, molecular weight is 90000 Da (predicted); Q8IU80-1: amino acid length is 802, molecular weight is 88874 Da (predicted); Q8IU80-2: amino acid length is 461, molecular weight is 51602 Da (predicted); Q8IU80-5: amino acid length is 824, molecular weight is 91333 Da (predicted).
The single-chain zymogen undergoes autoproteolytic processing. This results in TMPRSS6 shedding from the cell surface and conversion into an activated two-chains form which is released extracellularly. The process involves a trans-activation mechanism that requires TMPRSS6 oligomerization -
