Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), A549 (lane 2, 15 μg), HT-29 (lane 3, 15 μg), RAW264.7 (lane 4, 15 μg), and HaCaT (lane 5, 15 μg) using Toll-Like Receptor 5 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin(HRP), HY-P80955A, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C. Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Western blot analysis of extracts from Hela (lane 2(40μg),SK-Br-3 (lane 3(40μg), A549 (lane 4(40μg),U-251 (lane 5(40μg)using Toll-Like Receptor 5 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (1/1000)and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
Background
-
Function
Toll-Like Receptor 5 is a Pattern recognition receptor (PRR) located on the cell surface that participates in the activation of innate immunity and inflammatory response. Recognizes small molecular motifs named pathogen-associated molecular pattern (PAMPs) expressed by pathogens and microbe-associated molecular patterns (MAMPs) usually expressed by resident microbiota. Upon ligand binding such as bacterial flagellins, recruits intracellular adapter proteins MYD88 and TRIF leading to NF-kappa-B activation, cytokine secretion and induction of the inflammatory response. Plays thereby an important role in the relationship between the intestinal epithelium and enteric microbes and contributes to the gut microbiota composition throughout life[1][2][3][4][5].

Subcellular Localization
Cell membrane; Single-pass type I membrane protein
-
Expression
Tissue_specificity:Highly expressed on the basolateral surface of intestinal epithelia (PubMed:11489966) . Expressed also in other cells such as lung epithelial cells (PubMed:11489966, PubMed:18490781) -
Subunit
Homodimer (PubMed:22173220). Interacts with MYD88 (via TIR domain) (PubMed:20855887). Interacts with TICAM1 (via TIR domain) (PubMed:20855887).
