Romiplostim is an Fc-peptide fusion protein mimetic of TPO (thrombopoietin protein) and also a c-Mpl agonist. Romiplostim binds to c-Mpl, stimulates megakaryocyte-mediated thrombopoiesis, and activates the same downstream signaling pathways as endogenous TPO. Romiplostim is applicable to research related to hematopoietic acute radiation syndrome and chronic immune thrombocytopenia.

In Vivo

Romiplostim (3-300 μg/kg; s.c.; single dose at 24-26 h post-irradiation; daily on Days 1-3 post-irradiation; daily on Days 1-5 post-irradiation) provides a statistically significant ~40% increase in 30-day survival and hastens platelet recovery when administered as a single subcutaneous dose of 30 or 100 μg/kg 24 hours post-6.8 Gy total body irradiation, with no additional benefit from repeat dosing or co-administration with pegfilgrastim^[1].
