IL-4R alpha is the α subunit shared by the IL-4 and IL-13 receptors and was initially discovered in leukocytes. IL-4R alpha is coupled to the JAK1/2/3-STAT6 pathway and participates in promoting Th2 differentiation. IL-4R alpha binds to IL-4, causing phosphorylation of its C-terminal tyrosine residue. IL-4R alpha possesses an immunoreceptor tyrosine inhibitor motif (ITIM), and by phosphorylating ITIM, it can bind to the SH2 domain of a partial phosphatase. IL-4R alpha contains a transmembrane domain. IL-4R alpha protein, Canine is expressed by HEK293 cells and includes a type III fibronectin domain and a C-terminal C-6*His tag.

Biological Activity

Measured by its ability to inhibit IL-4-dependent proliferation of TF 1 human erythroleukemic cells. The ED50 for this effect is ≤18.24 ng/mL in the presence 0.2 ng/mL of Recombinant Human IL-4, corresponding to a specific activity is ≥5.482×104 units/mg.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
