Background

STX17, a pivotal member of the SNARE (soluble N-ethylmaleimide-sensitive factor-attachment protein receptor) family, assumes a crucial role in mediating the fusion of cellular membranes. This SNARE protein is localized on opposing membranes, where it assembles into a trans-SNARE complex—an extended, parallel four alpha-helical bundle that serves as the driving force behind membrane fusion. Specifically implicated in autophagy, STX17 exerts direct control over autophagosome membrane fusion with the lysosome membrane. Additionally, it may participate in the early secretory pathway, influencing the architecture of the endoplasmic reticulum-Golgi intermediate compartment/ERGIC and Golgi, and regulating transport between the endoplasmic reticulum, ERGIC, and Golgi. Within the context of autophagy, STX17 forms a SNARE complex alongside VAMP8 and SNAP29, orchestrating the fusion of autophagosomes with lysosomes.
