Description

Tirzepatide TFA (LY3298176 hydrochloride) is a glucose-dependent insulin nutritive polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor dual agonist. Tirzepatide TFA has neuroprotective effects and can be used in the study of type 2 diabetes, diabetes-related neuropathy and obesity[1][2][3].

IC50 & Target

Glucose-dependent insulinotropic polypeptide (GIP), GLP-1 receptor[1]

In Vitro

Tirzepatide TFA (0-0.4 μM, 7 days) is used to improve the neurodegeneration of SHSY5Y human neuroblastoma cells induced by high glucose and to overcome neuronal insulin resistance[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Western Blot Analysis[1]

Cell Line: SHSY5Y
Concentration: 0-0.4 μM
Incubation Time: 7 days
Result: Upregulated the mRNA expression and protein levels of both CREB and BDNF.
Increased the Bcl-2 and decreased BAX protein levels.
Increased the pAkt, MAP2, GAP43, and AGBL4 protein levels.
Increased GLUT4 and SORBS1 protein levels.
In Vivo

Tirzepatide TFA (10 nM/kg, subcutaneous injection) can inhibit colon cancer tumor growth in dietary-obese mice[2].
Tirzepatide TFA (1.35 mg/kg, intraperitoneal injection, once per week) improves spatial learning and memory impairment by modulating abnormal insulin resistance and inflammatory response in diabetic rats[3].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: Diet-induced obese (DIO) mice [2]
Dosage: 10 nM/kg
Administration: s.c.
Result: Reduced body weight and food consumption.
Reduced glucose, insulin, leptin, and c-peptide levels.
Animal Model: MC38 colon cancer mice model[2]
Dosage: 10 nM/kg
Administration: s.c.
Result: Reduced tumor volumes.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
