Background

BACE1, a member of the peptidase A1 family of aspartic proteases, plays a pivotal role in the pathogenesis of Alzheimer's disease. This gene gives rise to multiple transcript variants, one of which encodes a preproprotein undergoing proteolytic processing to generate the mature protease. Functioning as a transmembrane protease, BACE1 catalyzes the initial step in the formation of amyloid beta peptides from amyloid precursor protein. These peptides constitute the primary components of amyloid beta plaques, prominent in the brains of individuals with Alzheimer's disease. Homozygous knockout mice for BACE1 display a spectrum of nervous system defects, growth retardation, metabolic abnormalities, and elevated neonatal lethality. The broad expression of BACE1 across various tissues, including the cerebellum and ovary, underscores its significance beyond the central nervous system.

Biological Activity

Measured by its ability to cleave a fluorogenic peptide substrate, Mca-SEVNLDAEFRK(Dpn)RR-NH2. Read at excitation and emission wavelengths of 320 nm and 405 nm. The specific activity is 1035.061 pmoL/min/µg, as measured under the described conditions.
