It notably influences the activity of CREB1, MEF2D, JUN, and RORA, pivotal players in immune response, inflammation, and memory consolidation. In the thymus, CAMKIV regulates the selection threshold of CD4(+)/CD8(+) double positive thymocytes during T-cell ontogeny. In CD4 memory T-cells, it links T-cell antigen receptor (TCR) signaling to the production of IL2, IFNG, and IL4, achieved through the regulation of CREB and MEF2. CAMKIV further plays a role in the differentiation and survival phases of osteoclasts and dendritic cells (DCs), mediating DCs survival by connecting TLR4 signaling and the temporal expression of BCL2.
