ADC Control Human IgG1 (C214S light chain, C226V and C229V heavy chain)-Tesirine is an isotype control antibody-drug conjugate for MEDI3726 (HY-185729). It carries hinge mutations including C214S in the light chain and C226V/C229V in the heavy chain, and the toxic molecule-linker conjugate is Tesirine (HY-128952). ADC Control Human IgG1 (C214S light chain, C226V and C229V heavy chain)-Tesirine can be used in the research of prostate cancer and metastatic castration-resistant prostate cancer[1].

In Vivo

ADC Control Human IgG1 (C214S light chain, C226V and C229V heavy chain)-Tesirine (0.11-1 mg/kg; i.v.; single dose) induces a statistically significant increase in survival only at the highest tested dose of 1 mg/kg in male MF1 nude mice bearing LNCaP prostate cancer xenografts[1].

Animal Model: Hsd:Ola-MF1-Foxn1nu nude (4-8 week old male, 22.6-35.3 g, subcutaneous implantation of LNCaP cells)[1]
Dosage: 0.11 mg/kg; 0.33 mg/kg; 1 mg/kg
Administration: i.v.; single dose
Result: Increased survival statistically significantly at 1 mg/kg compared with control groups.
Showed tumor growth comparable to vehicle controls at 0.11 mg/kg and 0.33 mg/kg, with tumor volume trending similar to vehicle over the study period.
