Western blot analysis of extracts from 293T (lane2(20μg), RAW264.7 (lane3(20μg), Hela (lane4(20μg) and HepG2 (lane5(20μg) using Ferritin heavy chain Antibody (HY-P86448). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/3000) and Loading control antibody (Beta Actin, HY-P80993, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001 ,1/10,000) was used for 1 hour at room temperature.
Immunohistochemical analysis of paraffin-embedded human liver tissue using Ferritin heavy chain Antibody (YA6140). The section was pre-treated using heat mediated antigen retrieval with Tris/EDTA buffer (pH 9.0) for 20 minutes. The tissues were probed with the primary antibody (HY-P86448, 1/300) overnight at 4℃. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with neutral balsam.

Background

Function：Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity (PubMed:9003196). Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation (PubMed:9003196). Also plays a role in delivery of iron to cells (By similarity). Mediates iron uptake in capsule cells of the developing kidney (By similarity). Delivery to lysosomes is mediated by the cargo receptor NCOA4 for autophagic degradation and release of iron (PubMed:24695223, PubMed:26436293)

Subunit：Oligomer of 24 subunits (PubMed:9159481). There are two types of subunits: L (light) chain and H (heavy) chain (PubMed:9159481). The major chain can be light or heavy, depending on the species and tissue type. In the human liver, the heavy chain is predominant (PubMed:9159481). The functional molecule forms a roughly spherical shell with a diameter of 12 nm and contains a central cavity into which the insoluble mineral iron core is deposited (PubMed:9159481). Interacts with NCOA4; NCOA4 promotes targeting of the iron-binding ferritin complex to autolysosomes following starvation or iron depletion (PubMed:24695223)
