    Background

    The GST (Glutathione S-transferase) protein is integral to the cellular process of conjugating reduced glutathione to a diverse array of hydrophobic electrophiles, a crucial mechanism for detoxification and cellular defense.     Biological Activity

    Bio-activity is defined as the amount of enzyme that conjugate 1.0 μmole of 1-chloro-2,4-dinitrobenzene (CDNB) with reduced glutathione per minute at 37°C.

    Assay Procedure

    Materials
    Test protein: GST Protein, Schistosoma Japonicum (HY-P70270)
    Glutathione Transferase Activity Assay Kit

    Procedure
    1. Preheat the microplate reader for at least 30 minutes, setting the wavelength to 340 nm.
    2. Preheat selected reagents at 37°C for 15 minutes as required.
    3. Prepare sample group and blank group according to the following formula:
    (Note: Dilute GST protein to 0.05 mg/mL with ddH2O for use as sample)
    Sample group: 20 μL sample, 180 μL Reagent II, 20 μL Reagent III;
    Blank group: 20 μL Reagent I, 180 μL Reagent II, 20 μL Reagent III.
    Add the above reagents to 96-well UV plates, mix promptly by pipetting, and record the absorbance value A1 at 340 nm at 10 seconds (A1 blank). Incubate at 37°C for 5 minutes in a water bath (or set the microplate reader to 37°C if temperature-controlled). Remove, dry thoroughly, and measure absorbance at 340 nm at 5 minutes 10 seconds (A2 blank).
    4. Calculate enzyme activity based on protein concentration:
    Bio-activity is defined as the amount of enzyme that conjugate 1.0 μmole of 1-chloro-2,4-dinitrobenzene (CDNB) with reduced glutathione per minute at at 37°C.
