Carbonic Anhydrase 2 Protein, Human (C-His, Solution)

Carbonic Anhydrase 2 (CA2) catalyzes the reversible hydration of carbon dioxide. Mutations in CA2 are linked to osteopetrosis and renal tubular acidosis. The gene exhibits biased expression in various tissues, notably in the stomach and colon. Two transcript variants, encoding different isoforms, have been identified. Carbonic Anhydrase 2 Protein, Human (C-His, Solution) is the recombinant human-derived Carbonic Anhydrase 2 protein, expressed by E. coli , with C-His labeled tag.

Carbonic Anhydrase 2 Protein is a member of the carbonic anhydrase isozyme family, responsible for catalyzing the reversible hydration of carbon dioxide. Dysregulation of this enzyme is linked to conditions such as osteopetrosis and renal tubular acidosis. Two transcript variants encoding distinct isoforms have been identified. In addition to its fundamental role in carbon dioxide metabolism, the protein exhibits biased expression in various tissues, with notable levels in the stomach and colon, as well as eight other tissues. This tissue-specific expression profile suggests its potential involvement in specialized physiological processes beyond its well-established functions in acid-base balance.

Materials
Assay buffer: 12.5 mM Tris, 75 mM NaCl, pH 7.5
Test protein: Carbonic Anhydrase 2 Protein, Human (C-His, Solution) (HY-P72860)
Substrate: 4-Nitrophenyl acetate (4-NPA) , prepared as a 100 mM stock solution in ethanol.

Procedure
1.Dilute Human CA2 to 2 ng/μL in assay buffer.
2.Dilute the substrate to 2 mM in assay buffer.
3.Add 50 μL of 2 ng/μL Human CA2 to the wells, then add 50 μL of 2 mM substrate to initiate the reaction. Perform in triplicate.
4.Include a substrate blank with 50 μL of assay buffer and 50 μL of substrate, also in triplicate.
5.Read in kinetic mode at 400 nm for 5 minutes.
6.Calculate the specific activity:
