Background

The HA/Hemagglutinin protein binds to sialic acid-containing receptors on the cell surface, facilitating the attachment of the virus particle to the cell. This attachment leads to the internalization of approximately two thirds of the virus particles through clathrin-dependent endocytosis, while the remaining one third follows a clathrin- and caveolin-independent pathway. The protein plays a crucial role in determining the host range restriction and virulence. As a class I viral fusion protein, it is responsible for the penetration of the virus into the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane. In the low pH environment of endosomes, HA2 undergoes an irreversible conformational change, resulting in the release of the fusion hydrophobic peptide and the formation of a competent fusion pore. Multiple trimers of HA are necessary for the formation of this pore.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
