Background

The HA1/Hemagglutinin protein binds to receptors on the cell surface that contain sialic acid, facilitating the attachment of the virus particle to the cell. This attachment leads to internalization of approximately two thirds of the virus particles through a process called clathrin-dependent endocytosis, while about one third enters through a clathrin- and caveolin-independent pathway. The HA1 protein plays a crucial role in determining host range restriction and virulence of the virus. As a class I viral fusion protein, it enables the virus to penetrate the cell cytoplasm by mediating the fusion of the viral membrane with the endosomal membrane. The acidic environment in endosomes triggers a irreversible conformational change in the HA2 subunit, causing the release of a hydrophobic peptide required for fusion. The formation of a competent fusion pore necessitates the presence of multiple trimers of the HA1/Hemagglutinin protein.
