Background

The HA/Hemagglutinin Protein binds to sialic acid-containing receptors on the cell surface, facilitating attachment of the virus particle. This attachment leads to internalization of about two thirds of the virus particles through clathrin-dependent endocytosis and about one third through a clathrin- and caveolin-independent pathway. It plays a crucial role in determining host range restriction and virulence. As a class I viral fusion protein, it is responsible for penetrating the cell cytoplasm by mediating the fusion of the endocytosed virus particle membrane with the endosomal membrane. In the low pH environment of endosomes, there is an irreversible conformational change in HA2, which releases the fusion hydrophobic peptide. Multiple trimers of HA are necessary for the formation of a competent fusion pore.
