Background

The HA1 protein, also known as Hemagglutinin, binds to sialic acid-containing receptors on the cell surface, facilitating the attachment of the virus particle to the cell. This attachment leads to the internalization of about two thirds of the virus particles through clathrin-dependent endocytosis and approximately one third through a clathrin- and caveolin-independent pathway. HA1 also plays a crucial role in determining the host range restriction and virulence of the virus. As a class I viral fusion protein, it is responsible for penetrating the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane. The low pH environment in endosomes triggers an irreversible conformational change in HA2, releasing the fusion hydrophobic peptide. The formation of a competent fusion pore requires multiple trimers of HA.
