Leukotriene A4 Hydrolase/LTA4H Protein, Mouse (sf9, His)

Cat. No.: HY-P74776
Handling Instructions

LTA4H Protein, a bifunctional zinc metalloenzyme, functions as an epoxide hydrolase, converting leukotriene A4 (LTA4) to pro-inflammatory leukotriene B4 (LTB4). It also acts as an aminopeptidase, cleaving N-terminal arginines and countering inflammation, particularly through Pro-Gly-Pro (PGP) cleavage. LTA4H contributes to resolvin E1 and 18S-resolvin E1 biosynthesis, generating potent anti-inflammatory and pro-resolving lipid mediators. Its multifaceted activities underscore its intricate role in regulating inflammatory processes and lipid mediator biosynthesis. Leukotriene A4 Hydrolase/LTA4H Protein, Mouse (sf9, His) is the recombinant mouse-derived Leukotriene A4 Hydrolase/LTA4H protein, expressed by Sf9 insect cells, with C-His labeled tag. The total length of Leukotriene A4 Hydrolase/LTA4H Protein, Mouse (sf9, His) is 611 a.a., with molecular weight of ~62 kDa.

Background

The Leukotriene A4 Hydrolase/LTA4H protein is a bifunctional zinc metalloenzyme with both epoxide hydrolase (EH) and aminopeptidase activities. Serving as an epoxide hydrolase, it catalyzes the conversion of leukotriene A4 (LTA4) to the pro-inflammatory mediator leukotriene B4 (LTB4). Additionally, LTA4H exhibits aminopeptidase activity, displaying a high affinity for N-terminal arginines in various synthetic tripeptides. Beyond its pro-inflammatory EH role, the protein may counteract inflammation through its aminopeptidase activity, which cleaves the neutrophil attractant tripeptide Pro-Gly-Pro (PGP)—a bioactive collagen fragment generated by matrix metalloproteinase-9 (MMP9) and prolylendopeptidase (PREPL). Moreover, LTA4H participates in the biosynthesis of resolvin E1 and 18S-resolvin E1 from eicosapentaenoic acid, contributing to the generation of lipid mediators with potent anti-inflammatory and pro-resolving actions. The multifaceted activities of LTA4H highlight its intricate involvement in the regulation of inflammatory processes and lipid mediator biosynthesis.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
