NA/Neuraminidase Protein, Influenza A H5N1 (H255Y, HEK293) is the recombinant Virus-derived NA/Neuraminidase protein, expressed by HEK293 , with tag free and H255Y mutation. It originates from cell lysates collected after expressing a DNA sequence encoding the Influenza A virus neuraminidase.

Neuraminidase (NA) is described as a receptor-destroying enzyme because it cleaves a terminal sialic acid from the cellular receptors. Whereby, NA facilitates viral invasion of the upper airways, facilitates virus release during virus budding, prevents self-aggregation and ensures the efficient spread of the progeny virus from cell to cell, and also plays a role in the determination of host range restriction on replication and virulence. NA/Neuraminidase Protein, Influenza A H5N1 (H255Y, HEK293) is the recombinant Virus-derived NA/Neuraminidase protein, expressed by HEK293 , with tag free and H255Y mutation. It originates from cell lysates collected after expressing a DNA sequence encoding the Influenza A virus neuraminidase.

Background

Neuraminidase (NA) is described as a receptor-destroying enzyme because it cleaves a terminal sialic acid from the cellular receptors. NA may facilitate viral invasion of the upper airways by cleaving the sialic acid moities on the mucin of the airway epithelial cells. NA catalyzes the removal of terminal sialic acid residues from viral and cellular glycoconjugates, cleaves off the terminal sialic acids on the glycosylated HA during virus budding to facilitate virus release and additionally helps virus spread through the circulation by further removing sialic acids from the cell surface. These cleavages prevent self-aggregation and ensure the efficient spread of the progeny virus from cell to cell or the infection would be limited to one round of replication. NA also plays a role in the determination of host range restriction on replication and virulence[1][2].

Biological Activity

Measured by its ability to cleave a fluorogenic substrate, 2’-(4-Methylumbelliferyl)-α-D-N-acetylneuraminic acid. One unit is defined as the amount of enzyme required to cleave 1 nmole of 2'-(4-Methylumbelliferyl)-α-D-N-acetylneuraminic acid per minute at pH 7.5 at 37°C.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O.

Storage & Stability

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
