WS5 (Menthane carboxamidoethyl acetate) is a cooling agent fragrance ingredient[1].

In Vitro

WS5 (Up to 5000 μg/plate) is not mutagenic in the Ames test using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537, and Escherichia coli strain WP2uvrA[1].
WS5 (1008.75 μg/mL; 4-24 h) is not mutagenic to MLA cells[1].
WS5 (Up to 2000 μg/mL) is non-clastogenic in the micronucleus test using human peripheral blood lymphocytes[1].
WS5 does not react with skin peptides in the in vitro direct peptide reactivity assay[1].

WS5 (25-675 mg/kg; p.o.; daily; 91 days) has a no observed adverse effect level (NOAEL) for subchronic toxicity in Crl:CD(SD) rats[1].

Result: Showed no mortality across all doses.
Showed no treatment-related effects on body weights, food consumption, body temperature, or ophthalmic findings at any dose.
Reduced hematocrit values in males at 225 mg/kg/day, with no other treatment-related adverse effects.
Induced statistically significant increases in neutrophils, monocytes, and white blood cells in both sexes at 675 mg/kg/day.
Reduced hematocrit, hemoglobin, red blood cell counts, and mean corpuscular volume, plus increased red blood cell distribution width in both sexes at 675 mg/kg.
Increased serum creatinine, urea nitrogen, calcium, and triglyceride levels, plus total urine volume in both sexes at 675 mg/kg/day.
Decreased serum albumin/globulin ratios, chloride levels, and urine specific gravity in both sexes at 675 mg/kg.
Caused enlarged rough-surfaced kidneys in 1/10 males and pale kidneys in 1/10 females, plus increased kidney and liver weights at 675 mg/kg.
Induced kidney tubular degeneration, regeneration, dilatation, hyaline droplets, interstitial fibrosis, and tubular epithelium vacuolation in both sexes at 675 mg/kg/day.
Induced periportal hepatocellular vacuolation and centrilobular hepatocellular hypertrophy in both sexes at 675 mg/kg/day.
Induced cardiomyopathy in females at 675 mg/kg.
Showed slightly higher mean severity scores of α-2u-globulin nephropathy staining in male treatment groups, a species- and sex-specific effect not relevant to human health.
