Background

Beta-catenin, a pivotal player in the canonical Wnt signaling pathway, dynamically regulates cellular processes in response to Wnt ligands. In the absence of Wnt, beta-catenin participates in a complex with AXIN1, AXIN2, APC, CSNK1A1, and GSK3B, promoting its phosphorylation and ubiquitination, leading to subsequent degradation by the proteasome. Upon Wnt activation, beta-catenin accumulates in the nucleus, acting as a coactivator for transcription factors of the TCF/LEF family, thereby activating Wnt-responsive genes. This multifaceted protein also plays a role in cell adhesion, acting as a component of an E-cadherin:catenin adhesion complex. Beyond these functions, beta-catenin emerges as a negative regulator of centrosome cohesion and exhibits roles in insulin internalization, inhibition of anoikis, and promotion of neurogenesis.
