Background

CCN1/Cyr61 protein plays a multifaceted role in cellular processes, actively promoting cell proliferation, chemotaxis, angiogenesis, and adhesion. It demonstrates a crucial involvement in wound healing by orchestrating the up-regulation of key genes related to angiogenesis, inflammation, and matrix remodeling in skin fibroblasts. CCN1's gene-regulatory effects are heparin-binding dependent. Additionally, it exerts regulatory control by down-regulating the expression of collagen type-1 subunits alpha-1 and alpha-2. The protein facilitates cell adhesion and signaling through integrin alpha-6/beta-1, promotes cell migration via integrin alpha-v/beta-5, and stimulates cell proliferation through integrin alpha-v/beta-3. Its interactions with integrins are context-dependent, relying on heparin and the specific cell type. In skin fibroblasts, CCN1 binds ITGA6/ITGB1, in endothelial cells it binds ITGAV/ITGB3, and in platelets, it associates with ITGA2B/ITGB3, thereby influencing diverse cellular functions.

Biological Activity

Measured by its ability to mediate Balb/3T3 mouse embryonic fibroblast cell adhesion. Recombinant Human CCN1 immobilized at 0.1 μg/mL, 100 μL/well, will mediate 19.19% BALB/3T3 cell adhesion (3 x 104 cell/well added).

Reconstitution
