Background

Myeloperoxidase (MPO) protein plays a vital role in the host defense system of polymorphonuclear leukocytes, contributing to microbicidal activity against a diverse range of organisms. Upon stimulation of polymorphonuclear leukocytes, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid under physiological conditions, along with other toxic intermediates that significantly enhance microbicidal activity in these cells. Beyond its antimicrobial functions, MPO is involved in mediating the proteolytic cleavage of alpha-1-microglobulin, generating t-alpha-1-microglobulin. This processed form exhibits potent inhibitory effects on the oxidation of low-density lipoprotein particles, thereby contributing to the limitation of vascular damage.

Biological Activity

Measured by its ability to oxidize guaiacol in the presence of hydrogen peroxide., The specific activity is 16375.8504 pmol/min/µg, as measured under the described conditions.

Assay Procedure

Materials
Assay buffer: 20 mM MOPS, 0.1 M NaCl, 1 mM CaCl2, pH 7.0
Test protein: Myeloperoxidase/MPO Protein, Mouse (HEK293, His) (HY-P70282)
Hydrogen Peroxide Solution, 30% (v/v) (H2O2)
Guaiacol(HY-N1380)

Procedure
1. Dilute guaiacol to 100 mM in assay buffer containing 0.0034% H2O2 to prepare the substrate mixture. Mix thoroughly by shaking or stirring at room temperature for 15 minutes. Protect from light.
2. Dilute MPO to 3.34 μg/mL in assay buffer.
3. Dilute hydrogen peroxide from 30% to 0.00667% in assay buffer.
4. In a transparent 96-well UV plate, dispense 100 μL of 3.34 μg/mL Mouse MPO, followed by 100 μL of the diluted guaiacol/H2O2 mixture to initiate the reaction. Mix 100 μL of assay buffer with 100 μL of the diluted guaiacol/H2O2 mixture as the substrate blank.
5. Read in kinetic mode at 470 nm for 5 minutes. Use only the data from the first 10 seconds for calculations.
6. Calculate Specific Activity:
