Verified Bioactivity

Defined as the amount of hydroperoxide that 1μg of PRDX4 can reduce at 25°C for 1 minute. The specific activity is 1875- 2063.33 pmol/min/μg, as measured under the described conditions.

Assay Procedure

Materials
30% H2O2
PRDX4 Protein, Human (His) (HY-P71232)

Procedure
1. Dilute 30% hydrogen peroxide to concentrations of 0.3058, 0.1529, 0.076, 0.038, 0.019, 0.0096, 0.0047, 0 mol/L. Add 100 μL/well and measure the absorbance at 240 nm.
2. Dilute H2O2 to 0.2 mol/L.
3. Dilute the Human PRDX4 to 1000 μg/mL.
4. In a 96-well plate, add 90 μL of 0.2 mol/L H2O2 and 10 μL of 1000 μg/mL protein.
5. Read the initial absorbance at 240 nm.
6. Incubate at room temperature for 20 minutes and read the absorbance again.
7. Calculate specific activity:
