Its Fmoc group protects the α-amino group, and the Trt group protects the imidazole ring of the histidine side chain, reducing racemization and byproduct formation during synthesis. Fmoc-His(Trt)-OH can spontaneously self-assemble under conditions of pH < 4 to form a nanofiber network, constructing a hydrogel scaffold with a water content exceeding 99% (w/w). The fibers have a diameter of approximately 22 nm, matching the size of extracellular matrix (ECM) components[1].
