    PKM2 was a key enzyme regulated by ZEN to inhibit glycolysis and cell proliferation in goat endometrial epithelial cells. The NAD+/NADH ratio was altered. Cells were lysed in cold extraction buffer (10 min) and centrifuged (12 000g, 10 min, 4 °C), and supernatants were incubated (60 °C, 30 min) to hydrolyze NAD+. NAD+ and NADH concentrations were determined per the manufacturer's instructions, and their ratio was calculated.

This is achieved through colorimetric measurements based on the WST-8 color reaction, eliminating the need for purification of NAD+/NADH from the samples.

    • Do most proteins show cross-species activity?
