N-Methylprotoporphyrin IX (1 h) exhibits 17.8-fold and 33.3-fold lower binding affinities for the purified ferrochelatase ^P255R and ^P255G mutants, respectively, compared with wild-type ferrochelatase (the K_d values of the three are 0.16 ± 0.07 μM, 0.30 ± 0.10 μM, and 9 ± 5 nM, respectively)^[1].
N-Methylprotoporphyrin IX inhibits ferrochelatase with an apparent inhibition constant (K_i^app) of 3 nM; in contrast, the P255R and P255G mutants show significantly reduced sensitivity to NMPP, with corresponding K_i^app values of 1 μM and 2.3 μM, respectively^[1].
N-methylprotoporphyrin IX dimethyl ester (0.5-20 μM; up to 90 h) potently inhibits the synthesis of phycocyanin and chlorophyll a in C. caldarium cells cultured under light conditions^[2].
