Description

Previtamin D3 is an orally active, thermolabile steroidal compound that generates vitamin D3, as well as biologically inactive lumisterol and tachysterol. Previtamin D3 is synthesized in the skin of humans and rats via the UV-B-mediated photoconversion of 7-dehydrocholesterol. Previtamin D3 lacks direct anti-rachitic activity, and its in vivo activity relies on the vitamin D3 it produces. Previtamin D3 can be used in studies related to rickets and vitamin D metabolism[1][2].

In Vitro

Previtamin D3 undergoes temperature-dependent first-order interconversion with vitamin D3 in ethanol, with faster isomerization at higher temperatures[2].

Previtamin D3 (0.2 J/cm2; topical skin exposure; single dose) Exposure of vitamin-D-deficient rat skin converts cutaneous 7-dehydrocholesterol to previtamin D3, which does not thermally isomerize to vitamin D3 under low-temperature processing conditions[1].
Previtamin D3 (28-111 μg/1000 g consumed diet; p.o.; 7 doses at 3 to 4 day intervals) exhibits 35% of the antirachitic activity of pure vitamin D3 in rachitic North Holland Blue chicks, with this activity hypothesized to originate exclusively from vitamin D3 formed via in vivo isomerization[2].
