    The vaginal fibroblasts from women without POP were subjected to 0.6 μM H2O2 for 24 h in the presence and absence of a Wnt signaling inhibitor DKK-1 Protein, Human (HEK293, Fc) (DKK-1, 0.5 μg/mL; 24 h). Expression of collagen I, collagen III, MMP9 and TIMP1 protein in the vaginal fibroblasts derived from women without POP by Western blot. The results showed that ECM deposition caused by CSRNP1 knockdown was curbed after treatment with the Wnt pathway inhibitor DKK1.

    Adenovirus GFP-LC3 was used to detect the formation of autophagosome under Ang-II (1 μM), α-Klotho (400 pM), DKK-1 Protein, Human (HEK293, Fc) (DKK-1, 500 pM) and Wnt3a (1 nM) treatment for 72 h. Fluorescent puncta analysis exhibited that Klotho and DKK-1 promoted the formation of autophagosomes, as shown by an increase in green puncta; Klotho also relieved the inhibition of Wnt3a on autophagosome formation.

    Effects of crocin on the activation of WNT/β-catenin signaling during osteogenic differentiation of PDLSCs. PDLSCs were divided into three treatment groups as follows: Control, crocin, and crocin + DKK-1 Protein, Human (HEK293, Fc) (DKK1, 100 nM; 7 days). Western blotting was performed to determine the protein expression of β-catenin, c-Myc and cyclin D1 after 7 days of treatment. β-Actin served as the loading control. The results showed that supplementation with the Wnt pathway inhibitor DKK1 partially reversed the promotive effects of crocin on the protein expression of β-catenin and cyclin D1.

    Background

    DKK1 protein functions as a potent antagonist of canonical Wnt signaling through multiple mechanisms. It inhibits the interaction between LRP5/6 and Wnt and forms a ternary complex with the transmembrane protein KREMEN, facilitating the internalization of LRP5/6. Notably, DKK1 not only antagonizes the pro-apoptotic function of KREMEN1 in a Wnt-independent manner but also exhibits anti-apoptotic activity. The protein is implicated in limb development, where it modulates Wnt signaling to ensure normal limb patterning. Through its C-terminal Cys-rich domain, DKK1 interacts with LRP5 and LRP6, specifically engaging with beta-propeller regions 3 and 4 of LRP5. This interaction is further influenced by MESD and/or KREMEN, collectively leading to the attenuation of Wnt-mediated signaling. Additionally, DKK1 forms a ternary complex with LRP6 and KREM1, highlighting its multifaceted role in regulating crucial cellular processes and interactions with key proteins involved in Wnt signaling.

    1. Human LRP-6 (HY-P77990) immobilized on CM5 Chip can bind Human DKK-1 with an affinity constant of 1.574 nM as determined in a SPR assay.
    2. Measured by its ability to inhibit Wnt3a induced alkaline phosphatase production by C3H10T1/2 cells. The ED50 for this effect is approximately 0.2181 μg/mL in the presence of 10 ng/mL of Wnt3a, corresponding to a specific activity is 4585.053 units/mg.
