Background

GPR155 is a cholesterol-binding protein that functions as a regulator of the mTORC1 signaling pathway. It acts as a cholesterol sensor to modulate mTORC1 signaling: in the presence of cholesterol, GPR155 binds cholesterol, disrupting the interaction between the GATOR1 and KICSTOR complexes and promoting mTORC1 signaling; under cholesterol starvation, GPR155 fails to perturb the GATOR1-KICSTOR association, leading to mTORC1 inhibition. Additionally, GPR155 binds indole-3-acetic acid and may play a role in tryptophan metabolism. by similar: Studies further highlight GPR155's critical role in cholesterol sensing and mTORC1 regulation.
