PK3C3 Protein, Human (Active, sf9, His, GST)

The PI3KC3 protein is the catalytic subunit of the PI3K complex and mediates the formation of phosphatidylinositol 3-phosphate. PI3KC3-C1 initiates autophagosomes, whereas PI3KC3-C2 promotes autophagosome maturation and endocytosis. PK3C3 Protein, Human (sf9, His, GST) is the recombinant human-derived PK3C3 protein, expressed by sf9 insect cells , with N-8*His, N-GST labeled tag.

PK3C3 serves as the catalytic subunit of the PI3K complex, orchestrating the formation of phosphatidylinositol 3-phosphate. Various complex forms of PK3C3 play pivotal roles in diverse membrane trafficking pathways, with PI3KC3-C1 initiating autophagosomes and PI3KC3-C2 contributing to the maturation of autophagosomes and endocytosis. Within PI3KC3-C1, PK3C3 actively promotes the formation of endoplasmic reticulum membrane curvature before vesicle budding. Furthermore, PK3C3 is implicated in the regulation of degradative endocytic trafficking, essential for the abscission step in cytokinesis, likely in the context of PI3KC3-C2. Additionally, PK3C3 plays a crucial role in the transport of lysosomal enzyme precursors to lysosomes and is required for the progression from early to late endosomes. Notably, the kinase activity of PK3C3 is vital for the replication of SARS coronavirus-2 (SARS-CoV-2) during microbial infection.

The activity of PK3C3 is verified by the ADP-Glo method: Incubate PK3C3 protein, ATP, and the substrate at room temperature for 40 minutes, then add the ADP-Glo reagent and read the luminescence signal using the chemiluminescence module of the microplate reader. When the protein concentration is 1.5 nM, the conversion rate is about 20%.
