Calcein is a fluorescent dye and self-quenching probe, used as an indicator of lipid vesicle leakage, and also as a complexometric indicator for titration of calcium ions with EDTA, and for fluorometric determination of calcium. Calcein cannot directly cross the intact cell membrane of a living cell, unlike Calcein-AM (HY-D0041) which is cell-permeable. Calcein can also be used as a model drug for evaluating efficiency and bioavailability of drug delivery systems.

    Calcein is a fluorescent dye and self-quenching probe, used as an indicator of lipid vesicle leakage, and also as a complexometric indicator for titration of calcium ions with EDTA, and for fluorometric determination of calcium. Calcein cannot directly cross the intact cell membrane of a living cell, unlike Calcein-AM (HY-D0041) which is cell-permeable. Calcein can also be used as a model drug for evaluating efficiency and bioavailability of drug delivery systems[1][2].

    In Vitro

    Calcein is a fluorescent probe that emits yellow-green fluorescence and can be used to determine the labile iron pool as well as to measure the binding affinity of Fe(III) with ligands[3].
    Guide (The following is our recommended experimental protocol. This protocol is for reference only and should be modified according to your specific needs).
    1. Solution preparation[3]
    1.1 Preparation of stock solution
    Solvent: 1 M NaOH
    Concentration: 0.5 mM (optimize according to the experiment).
    Storage: Aliquot and store at -20°C or -80°C protected from light. Avoid repeated freeze-thaw cycles.
    1.2 Preparation of working solution
    Dilute to 0.5 μM with PBS or cell culture medium.
    Note: The working solution should be freshly prepared, used immediately, and protected from light.

    2. Cell staining
    2.1 Incubate cells (106/mL) with 20 μM Fe(II) at 37°C for 2 h.
    2.2 Collect cells by centrifugation (1000 rpm; 5 min), and stain by directly microinjecting Calcein (0.5 μM) into the cells (5-15 min).
    2.3 Wash cells twice with D'hanks buffer (pH 7.4).
    2.4 Monitor fluorescence using flow cytometry and confocal laser scanning microscopy.

    Coelomocytes suspensions (ISO and CF) are separately incubated with calcein-AM (excitation and emission wavelength: 496 nm and 516 nm, respectively) at a final concentration of 200 nM, during 30 min, at 26°C. Calcein-AM is a nonfluorescent ABC transporter substrate whose intracellular accumulation is inversely proportional to ABC transporter activity. Intracellular esterases convert calcein-AM into the fluorescent dye calcein, which is not an ABC transporter substrate, thereby accumulating the dye inside the cell. Therefore, a high fluorescence signal indicates low ABC transporter activity whereas a low fluorescence signal indicates high activity. The fluorescence of samples is measured by flow cytometer. The experiment is repeated six times in duplicates.
