Duvortuxizumab (MGD 011) is a chimeric humanized IG antibody targeting CD19 and CD3E^[1].

Duvortuxizumab is a dual-affinity re-targeting (DART) protein that binds to the CD3 molecule on T cells and the CD19 antigen on B cells. Duvortuxizumab exhibits good affinity to human CD3 and human CD19 with K_Ds of 21.2 nM and 2.0 nM^[2].
Duvortuxizumab exhibits cytotoxicity to CD19-positive cell Raji/GF (EC₅₀ is 0.02-0.03 ng/mL), causes depletion of malignant B cells in human PBMCs (EC₅₀ is 0.018 pg/mL)^[2].

  Flow cytometric analysis of 1×10⁶ Jurkat cells labelling CD3E (red) with Duvortuxizumab (HY-P99868). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control.
