Description

HKOCl-3 is a highly sensitive and selective fluorescent probe for detecting hypochlorous acid.Ex: 490 nm; Em 527 nm[1].

In Vitro

HKOCl-3 (10 μM) shows an increasing florescence intensity with increasing concentration of hypochlorous acid[1].
HKOCl-3 (10 μM) shows a much higher increase in fluorescence intensity of compound HKOCl-3 than treatment with other ROS (10 μM) and RNS (100 μM) [1].
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs)[2].
1. Solution preparation
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 1-2 mM (optimized according to the experiment).
1.2 Preparation of working solution
Dilute to 1-2 μM with with PBS or serum-free medium (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.

2. Cell staining
2.1 Cells are washed with PBS
2.2 Add HKOCl-3 (1 or 2 μM) in 1.5-mL HBSS supplemented with 0.1% Chloramphenicol (HY-B0239) and 0.6 mM L-Arginine (HY-N0455).
2.3 Cells are typically incubated for 30 min at 37°C.
2.4 Fluorescence microscopy detection (Ex/Em = 488/500-530 nm).
