Exosome-Specific 293 Cell Medium (Serum-Free) 

MCE Exosome-Specific 293 Cell Medium (Serum-Free) is specifically optimized for exosome production from HEK293 cells.

It features a chemically defined formulation, free of animal-derived components (Xeno-free), growth factors, and proteins. No serum supplementation is required during cell culture. When used in combination with the Exosome-Specific 293 Cell Medium Supplement (MCE Cat. No.: HY-K3114), it significantly enhances exosome secretion from 293 cells, achieving a 5-10 fold increase in yield compared to conventional HEK293 culture media.

Protect from light

1. Pre-warm the Exosome-Specific 293 Cell Medium (Serum-Free) to 37°C.

2. Prepare HEK293 cells in the logarithmic growth phase with ≥ 95% viability at a density of 2 × 10⁶ - 1 × 10⁷ cells/mL.

3. Inoculate the cells into the pre-warmed Exosome-Specific 293 Cell Medium (Serum-Free) at a seeding density of 3 × 10⁵ - 5 × 10⁵ cells/mL.

4. Culture the cells in sterile shake flasks or a bioreactor.

Shake flask culture conditions: 37°C, 80% humidity, 5%-10% CO₂, 140 rpm.

Bioreactor culture conditions: 37°C, pH 7.0, DO 40%, stirring speed 160 rpm.

5. Expand the HEK293 suspension cells until the density reaches 6 × 10⁶ - 8 × 10⁶ cells/mL.

Note: To further enhance exosome production, use the Exosome-Specific 293 Cell Medium Supplement (MCE Cat. No.: HY-K3114). Add the supplement at 25% of the culture volume. Continue culturing for 6-16 h, and collect the culture supernatant before cell viability drops below 90%.

6. The collected culture supernatant can be used for exosome isolation and purification.
