COA Handling Instructions

P-selectin is a Ca(2+)-dependent receptor on myeloid cells that specifically binds to carbohydrates on neutrophils and monocytes. It mediates interactions between activated endothelial cells or platelets and leukocytes by recognizing sialic acid-Lewis X.P-Selectin Protein, Mouse (HEK293, Fc) is the recombinant mouse-derived P-Selectin protein, expressed by HEK293 , with C-hFc labeled tag.

Description

P-selectin is a Ca(2+)-dependent receptor on myeloid cells that specifically binds to carbohydrates on neutrophils and monocytes. It mediates interactions between activated endothelial cells or platelets and leukocytes by recognizing sialic acid-Lewis X.P-Selectin Protein, Mouse (HEK293, Fc) is the recombinant mouse-derived P-Selectin protein, expressed by HEK293 , with C-hFc labeled tag.

Background

The P-selectin protein is a Ca(2+)-dependent receptor found on myeloid cells, which specifically binds to carbohydrates on neutrophils and monocytes. It plays a crucial role in mediating the interaction between activated endothelial cells or platelets and leukocytes. The ligand recognized by P-selectin is sialyl-Lewis X. P-selectin facilitates the rapid rolling of leukocytes over vascular surfaces during the initial stages of inflammation by interacting with SELPLG. It also interacts with SNX17. Moreover, P-selectin interacts with SELPLG/PSGL1 and PODXL2, promoting neutrophil adhesion and leukocyte rolling. This interaction requires the presence of the sialyl-Lewis X epitope on SELPLG and PODXL2, as well as specific tyrosine sulfation on SELPLG.

Biological Activity

Measured by the ability of the immobilized protein to support the adhesion of U937 human histiocytic lymphoma cells. The ED50 this effect is 5.013 μg/mL, corresponding to a specific activity is 199.48 units/mg.

Approximately 135 kDa

Purity
  • Greater than 85% as determined by reducing SDS-PAGE.
Appearance

Lyophilized powder.

Formulation

Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
