Skp2 inhibitor 2 (14ag) potently inhibits the Skp2-Cks1 protein-protein interaction in a cell-free HTRF assay with an IC50 of 0.57 μM[1].
Skp2 inhibitor 2 binds to the Skp2 protein via multiple hydrogen bonds and π-stacking interactions, occupying the Skp2-Cks1 binding site[1].
Skp2 inhibitor 2 (72 h) exhibits poor antiproliferative activity against Skp2-high-expressed MGC-803, NCI-H1299, MCF-7, EC-1, and PC-3 tumor cell lines, with IC50 values ranging from 51.8 μM to > 100 μM[1].
