    Description

    DiBAC4(3) is a voltage-sensitive fluorescent dyeex=490 nm, λem=505 nm). DiBAC4(3) can be used to measure cell membrane potential.

    In Vitro

    Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
    1. Solution preparation
    1.1 Preparation of stock solution
    Solvent: DMSO
    Concentration: 10-40 mM.
    Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
    1.2 Preparation of working solution
    Dilute to 10-40 μM with Hanks and 20 mM Hepes buffer (HHBS) or other buffer, such as pH 7 with 0.04% to 0.08% Poloxamer 407 (F127) (HY-D1005) and 2 mM Trypan Red (HY-D0983)).
    Note: The working solution should be prepared and used immediately. Keep it away from light.

    2. Cell staining
    2.1 Cells are plated in clear-bottomed black 96-well microplates.
    2.2 Add 100 μL DiBAC4(3) into the cell plate.
    2.3 Incubate the dye-loading plate in a cell incubator for 30 to 60 minutes.
    2.4 Run the membrane potential assay by monitoring the fluorescence intensity at Ex/Em = 490/505 nm.

    Prior to the fluorescence measurements, cells are incubated in KRH (Krebs-Ringer-HEPES) buffer containing with 100 nM DiBAC4(3) for 20 min at room temperature. The stained cells are used for experiments without washing. The fluorescence emission is collected using a 505 nm dicroic mirror and a BA filter (>520 nm)[1].
