Western blot analysis was performed on extracts from Hela (lane 1, 20 μg), A549 (lane 2, 20 μg), U2OS (lane 3, 20 μg), and K562 (lane 4, 20 μg) using LINE 1 ORF1p Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80955, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit/Mouse IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Background

Function：Nucleic acid-binding protein which is essential for retrotransposition of LINE-1 elements in the genome. Functions as a nucleic acid chaperone binding its own transcript and therefore preferentially mobilizing the transcript from which they are encoded

Subcellular Localization：Nucleus, nucleolus; Cytoplasm; Cytoplasm, Cytoplasmic ribonucleoprotein granule; Cytoplasm, Stress granule

Subunit：Homotrimer (via coiled coil domain). May also form larger homooligomers. May interact with DDX39A, HNRNPA1, SERBP1 and YBX1. Interacts with TEX19 and UBR2 (PubMed:28806172). Interacts with MOV10 (PubMed:23093941). Interacts with APOBEC3D; this interaction inhibits LINE-1 retrotransposition (PubMed:27428332)
