In Vitro

Cell Viability Assay[1]

Cell Line: Human B-cell acute lymphoblastic leukemia (B-ALL), acute myeloid leukemia (AML), T-cell acute lymphoblastic leukemia (T-ALL), diffuse large B-cell lymphoma (DLBCL) cell lines
Concentration: 10-100 ng/mL
Incubation Time: 48 h, 72 h
Result: Induced extensive cell death of human B-ALL, AML, T-ALL, and DLBCL cell lines after 48 and 72 hours.
Was significantly more effective at killing these blood cancer cells at 10-100 ng/mL than an αTIM-3 antibody at equivalent concentrations.
In Vivo

LYT-200 (1.5 mg/kg; weekly) administered at 1.5 mg/kg weekly provides survival benefit in immunocompromised mice with T-ALL comparable to methotrexate, and combination treatment with methotrexate yields the best survival outcomes in these models[1].
LYT-200 (1.5 mg/kg; weekly) administered at 1.5 mg/kg weekly provides superior survival protection compared to cytarabine in immunocompromised mice with AML, and combination treatment with cytarabine yields greater than 80% survival at 2 months post-disease appearance[1].

Biological Activity
  • Flow cytometric analysis of 1×106 Jurkat cells labelling Galectin-9 (red) with LYT-200 (HY-P991228). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa Isotype Control (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
  • Immobilized Galectin-9/LGALS9 Protein, Human (HEK293, His) can bind LYT-200. The EC50 for this effect is 40.78 ng/mL.
