IL-12 Protein, Mouse (HEK293)

Cat. No.: HY-P70666
COA Handling Instructions

IL-12 protein is a immune-suppressive heterodimeric cytokine, composed by IL-12A subunit (IL-12p35) and IL-12B subunit (IL-12p40), is naturally produced by dendritic cells. IL-12 exerts functions to activate and link the innate and acquired immune responses. IL-12 Protein, Mouse is produced in HEK293 cells, with total length of 506 amino acids and tag free.

Description

IL-12 protein is a immune-suppressive heterodimeric cytokine, composed by IL-12A subunit (IL-12p35) and IL-12B subunit (IL-12p40), is naturally produced by dendritic cells[1]. IL-12 exerts functions to activate and link the innate and acquired immune responses[2]. IL-12 Protein, Mouse is produced in HEK293 cells, with total length of 506 amino acids and tag free.

Background

IL-12 is primarily produced by professional antigen-presenting cells (APCs) such as B-cells and dendritic cells (DCs) as well as macrophages and granulocytes and regulates T-cell and natural killer-cell responses[2]
IL-12 belongs to the family of interleukin-12, the only heterodimeric cytokines, including IL-12, IL-23, IL-27 and IL-35[3]
IL-12 acts function by binding a receptor composed of IL12R1 and IL12R2 subunits, and results in the rapid tyrosine phosphorylation of a number of cellular substrates including the JAK family kinases TYK2 and JAK2[4]
IL-12 is an important factor for the differentiation of naive T cells into T-helper type 1 CD4+ lymphocytes secreting interferon-gamma[5]
IL-12 induces the production of interferon-gamma (IFN-gamma), favors the differentiation of T-helper 1 (Th1) cells and is an important link between innate resistance and adaptive immunity[2]

Biological Activity

Measured in a cell proliferation assay using CTLL-2 cells. The ED50 for this effect is 0.004776 ng/mL, corresponding to a specific activity is 2.09×108 units/mg.

20-25&38-45 kDa

Purity
  • Greater than 95% as determined by reducing SDS-PAGE.
Appearance

Lyophilized powder

Formulation

Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4 or 20 mM PB, 150 mM NaCl, pH 7.4.

Endotoxin Level

<1 EU/μg, determined by LAL method.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Reconstitution Calculator

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
