IL-12 protein is a immune-suppressive heterodimeric cytokine, composed by IL-12A subunit (IL-12p35) and IL-12B subunit (IL-12p40), is naturally produced by dendritic cells[1]. Background

IL-12 is primarily produced by professional antigen-presenting cells (APCs) such as B-cells and dendritic cells (DCs) as well as macrophages and granulocytes and regulates T-cell and natural killer-cell responses[2].
IL-12 belongs to the family of interleukin-12, the only heterodimeric cytokines, including IL-12, IL-23, IL-27 and IL-35[3].
IL-12 acts function by binding a receptor composed of IL12R1 and IL12R2 subunits, and results in the rapid tyrosine phosphorylation of a number of cellular substrates including the JAK family kinases TYK2 and JAK2[4].
IL-12 is an important factor for the differentiation of naive T cells into T-helper type 1 CD4+ lymphocytes secreting interferon-gamma[5].
IL-12 induces the production of interferon-gamma (IFN-gamma), favors the differentiation of T-helper 1 (Th1) cells and is an important link between innate resistance and adaptive immunity[2].

Verified Bioactivity

1.Measured in a cell proliferation assay using CTLL-2 cells. The ED50 for this effect is 0.004776 ng/mL, corresponding to a specific activity is 2.09×108 units/mg.
2.Measured by its ability to enhance IFN-gamma secretion in NK-92 human natural killer lymphoma cells. The ED50 for this effect is 2-10 ng/mL.
