[Abstract]; Targeted O-GlcNAcylation of CK2α by OGTAC-1 (100 nM) for 8 h significantly increased CK2α-CRBN interaction. Flag-CRBN, HTN-CK2α (WT or S347A), and fOGT were overexpressed in cells. HaloTrap was used for IP, and anti-FlagTag was used for co-IP detection.

Western blot analysis of protein Human SIRT5(lane 2(10ng) and lane 3(20ng)) and Human BRD4(lane 4(10ng) and lane 5(20ng)) using Flag Tag (HY-P80111) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

Function：FLAG Tag is a multifunctional fusion tag for the purification of recombinant proteins. FLAG Tag maintances the natural folding of its fusing proteins. FLAG Tag can be removed by enterokinase, and eluted under non-denaturing conditions.
