Octanoylcarnitine is an orally active medium-chain acylcarnitine transport intermediate in fatty acid β-oxidation. Octanoylcarnitine is converted to octanoyl-CoA by carnitine acetyltransferase (CrAT), which then generates energy via β-oxidation in mitochondria of the heart and skeletal muscle. Octanoylcarnitine enhances grip strength and treadmill endurance, alleviates lactic acidosis, distributes in muscle and heart tissues, increases free carnitine levels, and mitigates mitochondrial stress. Octanoylcarnitine is associated with long-chain fatty acid metabolism, shows a positive correlation with subcutaneous fat area in patients with metastatic pancreatic ductal adenocarcinoma, and is closely related to central retinal artery occlusion (CRAO).

Description

Octanoylcarnitine is an orally active medium-chain acylcarnitine transport intermediate in fatty acid β-oxidation. Octanoylcarnitine is converted to octanoyl-CoA by carnitine acetyltransferase (CrAT), which then generates energy via β-oxidation in mitochondria of the heart and skeletal muscle. In Vitro

Octanoylcarnitine (50 μM) maintains robust respiration to generate energy in isolated wild-type mouse skeletal muscle mitochondria[1].
Octanoylcarnitine (25 μM; 1 min) can be converted to octanoyl-CoA by recombinant human CrAT and CPT2 in vitro, exhibiting enzymatic activity[1].
Octanoylcarnitine (50 μM; 1 h) is efficiently converted to octanoyl-CoA by homogenates of wild-type mouse skeletal muscle and heart after incubation at 37℃[1].

Octanoylcarnitine (0.5 mg/g or 3% w/w; single intragastric administration or ad libitum intake for 10 days) improves exercise capacity, alleviates post-exercise lactic acidosis, and increases short-chain acylcarnitine levels in tissues and serum of LCAD-KO mice[1].
Octanoylcarnitine (0.5 mg/g; i.g.; single administration) significantly improves treadmill exercise capacity and grip strength in muscle-specific CPT2-deficient mice[1].
Octanoylcarnitine (0.5 mg/g; i.g.; single administration) is rapidly metabolized to CO2 in healthy mice, and the metabolite is detectable in exhaled breath within 10 min[1].

Animal Model: Long-Chain Acyl-CoA Dehydrogenase Knockout (LCAD-KO) (male, 12-16 weeks, LC-FAOD model)[1]
