Maridebart  (Synonyms: AMG133 antibody)

Maridebart (AMG133 antibody) is a human immunoglobulin G1-kappa, anti-GIPR (gastric inhibitory polypeptide receptor) monoclonal antibody. Maridebart is the antibody portion of Maridebart cafraglutide (HY-164535) and can be used for the synthesis of antibody-drug conjugates (ADC). Maridebart can be used for the reserrch of obesity and type 2 diabetes ^[1][2][3].

Maridebart's long-acting peptide-antibody conjugate-Maridebart cafraglutide (HY-164535) combines GLP-1 receptor agonist with glucose-dependent insulinotropic polypeptide (GIP) receptor antagonism. Maridebart cafraglutide shows antagonist activity against human, cynomolgus monkey and rat GIPR with IC₅₀ values of 46.4 nM, 26.5 nM, 822.3 nM, respectively. Maridebart cafraglutide shows agonist activity against human, cynomolgus monkey, rat and mouse GLP-1R with EC₅₀ values of 24.4 pM, 5.7 pM, 2.4 pM and 123 pM, respectively^[2].

  Immobilized Hu/cyno-GIPR-ECD-His can bind Maridebart. The EC₅₀ for this effect is 36.76 ng/mL.
• 
  Flow cytometric analysis of 1X10⁶ THP-1 cells with Maridebart (HY-P990047, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
