Metalaxyl-M ((R)-Metalaxyl) is an orally active and selective inhibitor of fungal RNA polymerase, which exerts fungicidal activity by selectively interfering with the synthesis of fungal ribosomal RNA. Metalaxyl-M can also be used to induce inflammation in hepatocytes and regulate tryptophan metabolism. Metalaxyl-M can be used in ecotoxicology studies[1][2][3].

In Vivo

Metalaxyl-M (200 mg/L; water exposure; daily water change; 23 days) significantly reduces the survival rate of earthworms (Tubifex tubifex), increases the rate of self-mutilation, and inhibits the activities of CAT, SOD, and GST enzymes[2].
Metalaxyl-M (10-60 mg/kg; oral administration; once a day; 28 days) induces hepatocyte inflammation, necrosis, and vacuolization in ICR male mice, interfered with urine metabolites and serum amino acid levels, and affects the tryptophan metabolic pathway[3].

Animal Model: ICR mice (male, 8-week-old) + mammalian toxicology model[3]
Dosage: 10 mg/kg; 30 mg/kg; 60 mg/kg
Administration: Oral gavage, once daily for 28 days
Result: Caused hepatocellular inflammation, necrosis, and vacuolation in liver, with 60 mg/kg showing severe damage.
Increased serum amino acids (serine, tyrosine, tryptophan) increased, while methionine and glutamine decreased.
Increased serotonin and indole-3-propionic acid (IPA), decreased indole-3-lactic acid (ILA) and kynurenine (KYN).
