1,3-Butanediol, an orally active ethanol dimer providing a source of calories for human nutrition. 1,3-Butanediol administeration can stimulate the biosynthesis of the ketone body, β-hydroxybutyrate (βHB). 1,3-Butanediol has cerebral protective and hypoglycaemic effect[1][2][3].

体内実験

1,3-Butanediol (25 mmol/kg, i.p.; 30 min prior to ischemia) improves neurologic deficit and reduces neuronal damage in the striatum and cortex of the hippocampus in cerebral ischemia rat model[1].
1,3-Butanediol (20%, in the drinking water, 4 weeks) increases systemic βHB concentration to a level similar as a 24-hour fast in rats[3].
1,3-Butanediol (5%, in the drinking water, 4 weeks) reverses age-associated endothelium-dependent and -independent vascular dysfunction in old rats[4].
1,3-Butanediol (25 mmol/kg, i.p., every 3 hours for 4 times) increases the plasma level of 3-hydroxybutyrate in rats, associated with a sustained 20% decrease in the plasma level of glucose[5].

To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    90% (20% SBE-β-CD in Saline)

    Solubility: ≥ 2.5 mg/mL (27.74 mM); Clear solution

    This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

    Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
  • Protocol 2

    Add each solvent one by one:  10% DMSO    90% Corn Oil

    Solubility: ≥ 2.5 mg/mL (27.74 mM); Suspended solution

    This protocol yields a suspended solution of ≥ 2.5 mg/mL (saturation unknown). Suspended solution can be used for oral and intraperitoneal injection.

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
