The vascular endothelial growth factor (VEGF) family of receptors (VEGFR) consists of three protein-tyrosine kinases (VEGFR1, VEGFR2, and VEGFR3) and two non-protein kinase co-receptors (neuropilin-1 and neuropilin-2). VEGFRs are typical tyrosine kinase receptors (TKRs) consist of an extracellular component with seven immunoglobulin-like domains, a single transmembrane domain, and a split tyrosine kinase domain. VEGF-A, -B, and PlGF bind to VEGFR-1; VEGFR-2 is stimulated by VEGF-A, -C, -D and -E; VEGFR-3 is specific for VEGF-C and -D. According to the consensus model for ligand-induced activation of RTKs, VEGF binds to a cognate VEGFR to induce receptor dimerization, leading to protein kinase activation and autophosphorylation of tyrosine residues in the receptor intracellular domains, and initiation of signaling pathways, such as PI3K/AKT and MAPK/ERK1/2. VEGFR signaling is tightly regulated at numerous different levels, including receptor expression levels, the availability and affinities for binding of its different ligands, the presence of VEGF-binding co-receptors, non-VEGF-binding auxiliary proteins and inactivating tyrosine phosphatases, the rate of receptor cellular uptake, extent of degradation and speed of recycling.
